For a traceable assessment, it helps to separate two questions: Which components can be distinguished in the sample? Do the measured properties match the expected molecule?

HPLC: Separation and chromatographic profile

High-performance liquid chromatography separates sample components under defined conditions. In UV analysis, detected components appear as peaks. A commonly used purity figure expresses the target peak area relative to the sum of the evaluated peak areas. Waters documents this calculation, for example, for a synthetic peptide at 220 nm.

Such a percentage always relates to the stated method. It is not a complete inventory of every conceivable impurity. Insufficiently separated components may also appear together. A distinctive new peak alone does not reveal its chemical identity.

LC-MS: Additional information about mass

LC-MS combines liquid chromatography with mass spectrometry. Alongside separation, it provides information about mass-to-charge ratios. The molecular mass derived from these can be compared with the expected mass. This supports identity testing and the investigation of by-products or changes. Which structural questions are actually answered depends on the measurement method and analysis; a matching mass alone does not prove every structural detail.

Question

Useful part of the report

Which peaks were separated?

Chromatogram and measurement conditions

How was purity calculated?

Detector, wavelength and integration rules

Does the measured mass match?

MS result comparing expected and measured values

How much peptide does the sample contain?

A separate content determination, if available

Why purity and content should be kept separate

Sigma-Aldrich explicitly distinguishes peptide purity from net peptide content. High purity therefore does not replace quantitative content determination. When comparing reports, it should be clear which measurement a figure refers to.

Frequently asked question: Is a single percentage enough?

For documentation, it is advisable to store the percentage, method, sample examined and result files together. If these details are missing, first request the complete report.

Further reading: HPLC basics and published laboratory reports.
